EVALUATION OF GROWTH OF Lactobacillus acidophilus IN TUMERIC (Curcuma longa) EXTRACT
Introdução
Lactobacillus acidophilus is a bacterium that has probiotic action, naturally present in the gastrointestinal tract, protecting the mucosa and aiding in food digestion. The objective of this study was to evaluate the prebiotic potential of the crude extract of Curcuma longa powder in the growth of Lactobacillus acidophilus.
Material e Métodos
The extraction was performed by weighing the turmeric powder in an Erlenmeyer flask and adding 80 % ethyl alcohol in a ratio of 1:3. After weighing, the Erlenmeyer flask was placed in an ultrasound bath for 50 minutes, at a frequency of 45 kHz, 50% power and a temperature of 40 ºC. The solid part was discarded and the liquid part was centrifuged at 5000 rpm for 20 minutes at 4ºC. The supernatant was rotary evaporated, sterilized in a 0.45 µm membrane and frozen until use. Lactobacillus acidophilus NCFM (FLORASTOR, União Química) was incubated in BHI broth at 37 ºC for 24 hours. Then, serial dilutions were performed until a cell suspension of 103 CFU/mL was obtained. In a 96-well ELISA plate, 100 µL of turmeric extract, 100 µL of BHI broth at double concentration and 100 µL of probiotic culture were added. The plate remained incubated for 24 hours at 37 ºC in a Multiskan skyhigh microplate spectrophotometer - Termo Fisher Scientific, readings were taken every hour at absorbance of 600 nm.
Resultados e Discussão
The results showed that in the first 8 hours Lactobacillus acidophilus in turmeric extract behaved like the positive control, 8 hours later the population grew above the control curve and continued to grow until the end of the 24 hours of incubation.
Conclusão
Therefore, Curcuma longa extract favored the growth of L. acidophilus confirming its prebiotic potential.
