Chemometric modeling of Piper corcovadensis C.DC leaves extract and evaluation of biological activities

Introdução

Piper corcovadensis C.DC is a plant found throughout South America, widely used in traditional medicine, and locally known as João Brandinho. This study aimed to determine the optimal conditions by a generalized linear model (GLM) for obtaining an ethanolic extract enriched with phenolic compounds from P. corcovadensis leaves, showcasing antioxidant and antitumor potential.

Material e Métodos

The effects were evaluated on cutaneous melanoma (SK-MEL-28), healthy peripheral blood mononuclear cells (PBMCs), reactive oxygen species (ROS), nitric oxide (NOX), and the endogenous defenses of protein (PSH) and non-protein thiols (NPHS). Exploration of solvent concentration factors (% water/ethanol), temperature (ºC), and extraction time (min) identified optimal conditions for extracting phenolic compounds with antioxidant activity. Total phenolic compounds (TPC) and antioxidant activity (AA) were quantified using Folin Ciocalteu and DPPH methods and analyzed by a generalized linear model (GLM), AA tests (ABTS and FRAP methods) supported the identified optimal extraction conditions. The biochemical markers of oxidative stress, intracellular antioxidant defenses, and cell viability were evaluated at 15.63, 62.50, and 250.00 µg mL-1 extract concentrations.

Resultados e Discussão

The optimal extraction condition, identified as treatment 8, involved an 80/20% ethanol/water ratio in a water bath at 80 ºC for 120 min. The resulting extract exhibited a TPC of 11.64±0.04 mg GAE g-1 and AA by the DPPH method of 21.27±0.535 mg mmol TE g-1. Additionally, AA by the ABTS and FRAP methods was 33.15±11.66 mmol TE g-1 and 13.47±1.37 mmol Fe2+ g-1, respectively. Notably, the extract demonstrated no cytotoxicity against healthy cells (PBMC) (p>0.0001) compared to the control. Although it did not induce cell death (p<0.0001) in tumor cells and had no significant impact on oxidative stress or antioxidant defenses, it is worth highlighting that the extract exhibited richness in phenolic compounds with antioxidant activity.

Conclusão

The extract from P. corcovadensis leaves remains a promising natural source of valuable antioxidant compounds, emphasizing its potential health benefits. Despite the extract's limited impact on certain cellular responses, its abundance in beneficial phenolic compounds suggests its relevance in the pursuit of natural antioxidants for health and well-being. Further research may unlock additional applications and insights into the therapeutic potential of P. corcovadensis.
